Does the MELO P1 support connecting an instrument and a speaker at the same time?

No, it does not. The MELO P1 features a single 3.5mm jack that can be used for either an instrument or a speaker.
After connecting your device via a 3.5mm cable, you must manually switch the 3.5mm Jack I/O Mode between Input or Output in the More Settings section of the HollyAudio App.
